Erythrocyte sedimentation rate (ESR or sed rate) is a test that indirectly measures the degree of inflammation present in the body. The test actually measures the rate of fall (sedimentation) of erythrocytes (red blood cells) in a sample of blood that has been placed into a tall, thin, vertical tube.

Green Top Tube. Phlebotomy tubes with green top contain sodium herapin, lithium herapin and ammonium herapin, all of which are coated in the inside of the tube. The herapin anticoagulant activates antithrombins that block the coagulation cascade and produce a plasma sample or whole blood.
Optimum results are from blood less than two hours old. The ESR is of limited diagnostic value in severe anemia or in hematologic states that affect increased size and shape variation (poikilocytosis) of the RBC (ie, presence of sickle cells or spherocytes).
