Between Chadbourn and Wilmington, U.S. 76 runs concurrent with U.S. 74, a distance of approximately . The highway's eastern terminus is Wrightsville Beach, North Carolina. Its western terminus is Chattanooga, Tennessee.

In Georgia, the highway traverses the northern part of the state and passes through the Chattahoochee National Forest and the Georgia's most mountainous region. From east to west, U.S. 76 passes through Rabun, Towns, Union, Fannin, Gilmer, Murray and Whitfield Counties.South Carolina
In South Carolina, U.S. 76 is a well-traveled route spotted with small towns (35 mph speed limit), factory entrances and municipalities. The highway parallels Interstate 26 to the east, thus serving as the main local route between the small industrial towns between Columbia and Anderson.
